Between fight and flight, the human beings have the freedom to choose. We have the unique capacity to decide the quality of our life, independent of the will of others. We need the support and cooperation of others, and their input will prove to be very important in deciding our success. But ultimately it is our free will which should decide our life.

Seeking help of others is different from seeking their permission. When we seek help, we still retain our freewill. On the other hand, when we seek their permission, we are likely to surrender our freewill. When we decide on something, we have our own reasons and justifications for our decision. Sometimes we may not be guided by logical reasoning, but our intuition. Others may not have this input. Hence, they may not agree with our view point. Sometimes we may not have the maturity to accept the decision of others when we ask their permission and in the process lose our relationship.

Empowerment is a result of our willingness to take charge of our life. When we take charge of our own life and fate, we may go through a lot of tension and stress. But that is the only way to exercise our freewill.

Over this weekend, you may do an exercise. Take two important decisions you took over the last six months and check whether you sought the help or the permission of others to decide, and notice what happened to the quality of your decision!

First quarter GDP revised slightly lower; austerity bites

WASHINGTON (Reuters) - A drop in government spending dragged more on the U.S. economy than initially thought in the first three months of the year, although consumer spending looked relatively resilient to Washington's austerity drive.

Other reports on Thursday showed the number of new jobless claims rose modestly last week while contracts on previously owned homes climbed to a three-year high in April.

Together, the reports pointed to an economy that has held up reasonably well despite government constraints, but nevertheless faced headwinds severe enough to dissuade the U.S. Federal Reserve from trimming its monetary stimulus in the immediate future.

"(The reports) paint the picture of an economy with strengthening fundamentals that is facing significant fiscal drag," said Ellen Zentner, an economist at Nomura in New York.

Gross domestic product, a measure of the country's total economic output, expanded at a 2.4 percent annual rate during the first quarter, down a tenth of a point from an initial estimate, the Commerce Department said.

Analysts had forecast a 2.5 percent gain.

Government spending tumbled at a 4.9 percent annual rate, which was faster than the 4.1 percent rate initially estimated. Also holding back growth during the quarter, businesses outside the farm sector stocked their shelves at a slower pace.

Washington has been tightening its belt for several years but ramped up austerity measures in 2013, hiking taxes in January and slashing the federal budget in March.

"We are dramatically under-spending in Washington," said Michael Strauss, a market strategist at Commonfund in Wilton, Connecticut.

U.S. stocks rose and the dollar weakened as some investors bet the data could dissuade the Fed from rushing to taper a bond buying program that has acted as a bulwark against government belt tightening. Prices for U.S. government debt pared losses.

Despite the signs of a substantial fiscal drag, the GDP report also highlighted a resilience that has surprised many economists.

Consumer spending, which accounts for more than two-thirds of U.S. economic activity. rose at a 3.4 percent annual rate, up two tenths of a point from the government's previous estimate.

Excluding the volatile inventories component, GDP rose at an upwardly revised 1.8 percent rate, slightly higher than analysts had forecast. This suggests that an improvement in hiring and incomes over the last year has helped keep economic momentum intact.

"(It's) steady as she goes," said Stephen Stanley, an economist at Pierpont Securities in Stamford, Connecticut

HOUSING RECOVERY

Most economists still expect economic growth will slow around the middle of 2013 as budget cuts are enacted. But growth is seen picking up by year end, propelled by consumer spending and an apparently entrenched housing market recovery.

In April, the National Association of Realtors' index of signed contracts for home resales rose 0.3 percent to 106.0, the highest reading since April 2010.

The housing market recovery is being driven by the Fed's very easy monetary policy stance, which has kept mortgage rates low. Although speculation the Fed could begin to curtail its bond buying within a few months has driven mortgage rates sharply higher in recent days, economists say the fundamentals of the housing recovery still appear strong.

A separate report showed the number of Americans filing new claims for unemployment benefits unexpectedly rose last week, but not enough to suggest a shift in the recent pattern of steady job gains.

Initial claims for state unemployment benefits increased 10,000 to a seasonally adjusted 354,000, above analysts' expectations, Labor Department data showed.

World stocks volatile amid growth uncertainty

BANGKOK (AP) ? Global stock markets were volatile Thursday, with the Japanese benchmark dropping sharply while those in Europe edged higher, as investors muddled through conflicting signals about the state of the global economy.

Questions about whether the U.S. Federal Reserve would begin to withdraw its monetary stimulus program were also a source of uncertainty for markets.

Japan's Nikkei plummeted more than 5 percent, with investors increasingly doubting the government's economic strategy can extricate the country from years of economic malaise. The yen rose against the dollar, generally regarded as a negative for Japan's powerhouse export sector.

European stocks gained ahead of a meeting between French President Francois Hollande and German Chancellor Angela Merkel in Paris, where they will discuss the introduction of a full banking union for Europe.

Britain's FTSE 100 rose 0.2 percent to 6,643.04. Germany's DAX was marginally higher at 8,339.23. France's CAC-40 advanced 0.4 percent to 3,988.36.

Wall Street appeared headed for a second day of losses, with Dow Jones industrial futures falling 0.2 percent to 15,262. S&P 500 futures declined 0.2 percent to 1,643.30.

Sharp rises in global stock markets this year have been partly fueled by central bank actions to keep interest rates super low to support economic recovery in the U.S., Europe and Japan.

Positive signs of growth in the U.S., including data released Tuesday showing improved consumer confidence and housing prices, have also helped to boost Wall Street stocks to record highs.

However, an improving U.S. economy increases the chance that the Fed might ease back on its massive bond-buying program, known as quantitative easing. The purchase of $85 billion a month in Treasury bonds has helped keep interest rates down and been a boon to stock markets, where investors have fled in search of higher returns.

Investment enthusiasm was also curbed by warnings from the Paris-based Organization for Economic Cooperation and Development, which said Wednesday that Europe's recession risked hurting the world economy. The OECD slashed its forecast for the combined economy of the 17 countries that use the euro, saying it will shrink by 0.6 percent this year, after a 0.5 percent drop in 2012.

"The fact that the OECD added to the gloom yesterday with a raft of downgrades to its growth forecasts has added to the murky economic picture," said Michael Hewson of CMC Markets in a commentary.

Japan's Nikkei 225 index tumbled 5.2 percent to close at 13,589.03. The Tokyo benchmark has shown stomach-churning volatility this month, plunging more than 3 percent on Monday and 7 percent on May 23. However, the Nikkei is up 30 percent for the year.

Hong Kong's Hang Seng shed 0.3 percent to 22,484.31. Australia's S&P/ASX 200 dropped 0.9 percent to 4,930.70. Benchmarks in Singapore, Taiwan, and Indonesia fell more than 1 percent. The Philippines dropped 3.8 percent. South Korea's Kospi was marginally lower at 2,000.10.

Daniel Martin of Capital Economics in Singapore said two concerns are generally weighing on emerging market stocks.

"The first is that the Fed might soon start tapering its assets purchases. The second is that growth in Asia has generally disappointed the market this year, as the global trade recovery has faltered," he said.

One particular pocket of concern is China. A survey by HSBC Corp. showed that manufacturing in the world's No. 2 economy slipped in May, a sign the country's fragile recovery might be weakening. The official monthly figure on factory output is due Saturday.

Otherwise, the lack of major data releases for the day deprives investors of reasons to wade into stocks, analysts said.

"A lack of first tier data releases today will limit activity although the tone will likely remain relatively downbeat," said Mitul Kotecha of Credit Agricole CIB in a market commentary.

The latest speculation surrounding the Fed came after the release of positive consumer confidence and housing news on Tuesday. That led investors to fret over the prospect of the Fed reducing its bond-buying.

Japanese export stocks fell as the yen crept higher against the dollar. Honda Motor Corp. fell 3.4 percent. Yamaha Motor Co. lost 3.6 percent.

Benchmark oil for July delivery was down 53 cents to $92.60 per barrel in electronic trading on the New York Mercantile Exchange. The contract for the benchmark grade fell $1.88 to close at $93.13 a barrel on the Nymex on Wednesday.

In currencies, the euro rose to $1.2979 from $1.2934 late Thursday in New York. The dollar fell to 100.77 yen from 101.15 yen.

Gene therapy gives mice broad protection to pandemic flu strains, including 1918 flu

May 29, 2013 ? Researchers at the Perelman School of Medicine, University of Pennsylvania have developed a new gene therapy to thwart a potential influenza pandemic. Specifically, investigators in the Gene Therapy Program, Department of Pathology and Laboratory Medicine, directed by James M. Wilson, MD, PhD, demonstrated that a single dose of an adeno-associated virus (AAV) expressing a broadly neutralizing flu antibody into the nasal passages of mice and ferrets gives them complete protection and substantial reductions in flu replication when exposed to lethal strains of H5N1 and H1N1 flu virus. These strains were isolated from samples associated from historic human pandemics -- one from the infamous 1918 flu pandemic and another from 2009.

Wilson, Anna Tretiakova, PhD, Senior Research Scientist, Maria P. Limberis, PhD, Research Assistant Professor, all from the Penn Gene Therapy Program, and colleagues published their findings online this week in Science Translational Medicine ahead of print. In addition to the Penn scientists, the international effort included colleagues from the Public Health Agency of Canada, Winnipeg; the University of Manitoba, Winnipeg; and the University of Pittsburgh. Tretiakova is also the director of translational research, and Limberis is the director of animal models core, both with the Gene Therapy Program.

"The experiments described in our paper provide critical proof-of-concept in animals about a technology platform that can be deployed in the setting of virtually any pandemic or biological attack for which a neutralizing antibody exists or can be easily isolated," says Wilson. "Further development of this approach for pandemic flu has taken on more urgency in light of the spreading infection in China of the lethal bird strain of H7N9 virus in humans."

At the Ready Influenza infections are the seventh leading cause of death in the United States and result in almost 500,000 deaths worldwide per year, according to the Centers for Disease Control. The emergence of a new influenza pandemic remains a threat that could result in a much loss of life and worldwide economic disruption.

There is also interest by the military in developing an off-the-shelf prophylactic vaccine should soldiers be exposed to weaponized strains of infectious agents in biologic warfare.

Human antibodies with broad neutralizing activity against various influenza strains exist but their direct use as a prophylactic treatment is impractical. Now, yearly flu vaccines are made by growing the flu virus in eggs. The viral envelope proteins on the exterior, namely hemagglutinin, are cleaved off and used as the vaccine, but vary from year to year, depending on what flu strains are prevalent. However, high mutation rates in the proteins result in the emergence of new viral types each year, which elude neutralization by preexisting antibodies in the body (specifically specific receptor binding sites on the virus that are the targets of neutralizing antibodies).

This approach has led to annual vaccinations against seasonal strains of flu viruses that are predicted to emerge during the upcoming season. Strains that arise outside of the human population, for example in domestic livestock, are distinct from those that normally circulate in humans, and can lead to deadly pandemics.

These strains are also not effectively controlled by vaccines developed to human strains, as with the 2009 H1N1 pandemic. The vaccine development time for that strain, and in general, was not fast enough to support vaccination in response to an emerging pandemic.

Knowing this, the Penn team proposed a novel approach that does not require the elicitation of an immune response, which does not provide sufficient breadth to be useful against any strain of flu other than the one for which it was designed, as with conventional approaches.

The Penn approach is to clone into a vector a gene that encodes an antibody that is effective against many strains of flu and to engineer cells that line the nasal passages to express this broadly neutralizing antibody, effectively establishing broad-based efficacy against a wide range of flu strains.

A Broad Approach The rational for targeting nasal epithelial cells for antibody expression was to focus this expression to the site of the body where the virus usually enters the body and replicates which is the nasal and oral mucosa. Antibodies are normally expressed from B lymphocytes so one challenge was to design vectors that could deliver antibody genes to the non- lymphoid respiratory cells of the nasal and lung passages and could express functional antibody protein.

Targeting the respiratory cells was achieved through the use of a vector based on a primate virus -- AAV9 -- which was discovered in the Wilson laboratory and evaluated previously by Limberis for possibly treating patients with cystic fibrosis. The team constructed a genetic payload for AAV9 that expressed an antibody that was showed by other investigators to have broad activity against flu.

Efficacy of the treatment was tested in mice that were exposed to lethal quantities of three strains of H5N1 and two strains of H1N1, all of which have been associated with historic human pandemics (including the infamous H1N1 1918).

Flu virus rapidly replicated in untreated animals all of which needed to be euthanized. However, pretreatment with the AAV9 vector virtually shut down virus replication and provided complete protection against all strains of flu in the treated animals. The efficacy of this approach was also demonstrated in ferrets, which provide a more authentic model of human pandemic flu infection.

"The novelty of this approach is that we're using AAV and we're delivering the prophylactic vaccine to the nose in a non-invasive manner, not a shot like conventional vaccines that passively transfer antibodies to the general circulation," says Limberis.

"There's a long history of using antibodies for cancer and autoimmune disease, but only two have been approved for infectious diseases," notes Tretikova. "This novel technique has allowed for the development of a prophylactic passive vaccine that is cost effective, easily administered, and quickly manufactured."

The team is working with various stakeholders to accelerate the development of this product for pandemic flu and to explore the potential of AAV vectors as generic delivery vehicles for countermeasures of biological and chemical weapons.

Regarding component shortages, it?s not always a production issue that causes problems in HTC?s supply chain and an interesting tidbit emerged earlier this week as?HTC president for the North Asian region,?Jack Tong, spoke to members of the press in Taiwan.

As HTC?s Jack Tong recounted his company?s troubles following the launch of the HTC Desire, he slipped in a pretty huge accusation. Tong said that the Desire initially launched with an AMOLED display supplied by Samsung. After the phone started gaining momentum and sales picked up, the executive says Samsung suddenly couldn?t supply it with panels anymore.

?We found that key component supply can be used as a competitive weapon,? Tong told reporters, according to Focus Taiwan.?HTC ended up having to redesign the Desire and relaunch it without the Samsung-built AMOLED displays.

Cosmic glitch: Astronomers discover new phenomenon in neutron star

May 29, 2013 ? The physics behind some of the most extraordinary stellar objects in the Universe just became even more puzzling.

A group of astronomers led by McGill researchers using NASA's Swift satellite have discovered a new kind of glitch in the cosmos, specifically in the rotation of a neutron star.

Neutron stars are among the densest objects in the observable universe; higher densities are found only in their close cousins, black holes. A typical neutron star packs as much mass as half-a-million Earths within a diameter of only about 20 kilometers. A teaspoonful of neutron star matter would weigh approximately 1 billion tons, roughly the same as 100 skyscrapers made of solid lead.

Neutron stars are known to rotate very rapidly, from a few revolutions per minute to as fast as several hundred times per second. A neutron star glitch is an event in which the star suddenly begins rotating faster. These sudden spin-up glitches have long been thought to demonstrate that these exotic ultra-dense stellar objects contain some form of liquid, likely a superfluid.

This new cosmic glitch was detected in a special kind of neutron star -- a magnetar -- an ultra-magnetized neutron star that can exhibit dramatic outbursts of X-rays, sometimes so strong they can affect Earth's atmosphere from clear across the galaxy. A magnetar's magnetic field is so strong that, if one were located at the distance of the Moon, it could wipe clean a credit card magnetic strip here on Earth.

Now astronomers led by a research group at McGill University have discovered a new phenomenon: they observed a magnetar suddenly rotate slower -- a cosmic braking act they've dubbed an "anti-glitch." The result is reported in the May 30 issue of Nature.

The magnetar in question, 1E 2259+586 located roughly 10,000 light years away in the constellation of Cassiopeia, was being monitored by the McGill group using the Swift X-ray telescope in order to study the star's rotation and try to detect the occasional giant X-ray explosions that are often seen from magnetars.

"I looked at the data and was shocked -- the neutron star had suddenly slowed down," says Rob Archibald, lead author and MSc student at McGill University. "These stars are not supposed to behave this way."

Accompanying the sudden slowdown, which rang in at one third of a part per million of the 7-second rotation rate, was a large increase in the X-ray output of the magnetar, telltale evidence of a major event inside or near the surface of the neutron star.

"We've seen huge X-ray explosions from magnetars before," says Victoria Kaspi, Professor of Physics at McGill and leader of the Swift magnetar monitoring program, "but an anti-glitch was quite a surprise. This is telling us something brand new about the insides of these amazing objects." In 2002, NASA's Rossi X-ray Timing Explorer satellite also saw a large X-ray outburst from the source, but in that case, it was accompanied by a more usual spin-up glitch.

The internal structure of neutron stars is a long-standing puzzle, as the matter inside these stars is subject to forces so intense that they are presently not re-creatable in terrestrial laboratories. The densities at the hearts of neutron stars are thought to be upwards of 10 times higher than in the atomic nucleus, far beyond what current theories of matter can describe.

The reported anti-glitch strongly suggests previously unrecognized behaviour inside neutron stars, possibly with pockets of superfluid rotating at different speeds. The researchers further point out in the Nature paper that some properties of conventional glitches have been noted to be puzzling and suggestive of flaws in the existing theory to explain them. They are hoping that the discovery of a new phenomenon will open the door to renewed progress in understanding neutron star interiors.

(AP) ? Several times around the start of these playoffs, Miami coach Erik Spoelstra insisted that the postseason path his team would wind up navigating had the potential to be more challenging than the route they took to the NBA championship a year ago.

He's apparently correct, probably to his own chagrin.

The defending NBA champion Heat are in a bit of trouble. They can't get enough rebounds, can't get Dwyane Wade on track, can't get consistency out of Chris Bosh ? and will likely see all those story lines either grow exponentially or basically disappear Thursday night, when they host to the Indiana Pacers in Game 5 of a super-competitive Eastern Conference finals that's now knotted at two games apiece.

"We have a great locker room of Alpha competitors," Spoelstra said Wednesday. "And so they take this very seriously. We're playing against a worthy opponent and if we don't play well, they beat us. If they don't play well and we impose our identity, we beat them. That's what this is all about. So let's lace 'em up and let's get ready for Game 5."

Game 6 will be in Indiana on Saturday night, while the Western Conference champion San Antonio Spurs keep waiting to see who they'll face in the NBA Finals starting on June 6.

History says the Game 5 winner when a series is tied at 2-2 has a colossal upper hand, though that's an axiom that the Heat both proved and disproved last season.

When the Heat and Pacers split the first four games of their second-round series last year, Miami rolled to a 115-83 home win in Game 5 and captured the series in six games. One round later, Miami lost a home Game 5 of the East finals to Boston, then went on the road for Game 6 and got a virtuoso 45-point night from LeBron James to stave off elimination before coming home and winning a nailbiter of a Game 7 to advance.

Given all that, it's no wonder why Spoelstra said the Heat aren't looking back at any series as a blueprint for how the final acts of this one should go.

"We don't need confidence to go into any game," James said. "We're a confident bunch. We're excited to get the opportunity to go back to our home and play Game 5."

Confidence is not exactly in short supply around the Pacers right now, either.

Indiana came into the series saying ? and believing ? that it could find a way to oust the team that was virtually preordained as a champion entering these playoffs. That hasn't changed.

"We've got to be at our best," Pacers coach Frank Vogel said Wednesday. "Our intensity, our determination, our focus, we've got to keep getting better if we're going to beat this team."

The statistical trends probably aren't surprising. The Heat have scored 402 points, the Pacers 394. The Heat have shot 47 percent from the field, the Pacers 46 percent. The Pacers have shot 37 percent from 3-point range, the Heat 34 percent.

Miami is better at forcing turnovers, the Pacers are better at rebounding. Miami has forced Indiana into 14 more turnovers in the series, but the Pacers are outrebounding the Heat by 10 boards per game. Pacers center Roy Hibbert is averaging 12 rebounds; Bosh has grabbed 13 rebounds ? total ? in the series, or as many as Miami guard Ray Allen has despite being half a foot shorter and playing 32 fewer minutes.

"We know what they run, they know what we run," Hibbert said. "So I guess it's more about who wants it more."

Hibbert has had no match in this series. Miami plays without a true center, and Bosh often ends up in the same general spots on offense as a guard would. So at 7-foot-2, Hibbert almost seems like he has permission to do whatever he wants in the paint, and has picked the right time to play the series of his life.

"We don't want to go home," Hibbert said. "Miami is one of the best teams, along with San Antonio, and we know we have an uphill battle and we trust each other and love each other and that's just grown, that love for each other. This is probably the first team I've felt like that about because last year even there were some guys were not all about winning."

That can't be said now.

"We understand that the champs are going to come back and be ready to play at home," Pacers star Paul George said. "And we need to be ready to come out and play well."

Game 5 was a cavalcade of mistakes for the Heat, who had their second-worst shooting night of the entire season at 39 percent.

James fouled out for only the fifth time in his career, though Miami was clearly miffed at some late-game calls. Bosh went 1 for 6 with only three rebounds. Wade, that aching right knee still apparently an issue, missed 10 of his 15 shots. And despite it all, the Heat still had the lead in the final minutes.

"We missed an opportunity to go up 3-1," Bosh said. "But we did get home-court back. We did our job. We have to come out and play with a lot of passion."

Wade, who's averaging an uncharacteristically low 13.4 points in his last 10 games, was more succinct: "Our whole team has to do more," he said.

Being in a 2-2 series isn't the right time for Heat panic over now being in what amounts to a best-of-three, or Heat lament over anything that isn't going their way, Spoelstra said. At one point Wednesday, he took his left fist and right fist, banged them together to make a point, and insisted that the series will simply be won by the team that does enough to deserve a chance at facing the Spurs for the title.

"Our guys have respect for the other team in that locker room," Spoelstra said. "We know nothing is earned easy in this series. We've got to work for it. Being home doesn't guarantee anything."

International crew takes short cut to space station

CAPE CANAVERAL, Florida (Reuters) - A Russian spaceship took a shortcut to the International Space Station on Tuesday, delivering a veteran cosmonaut, a rookie Italian astronaut and an American mother on her second flight to the outpost in less than six hours.

The capsule slipped into its berthing port at 10:10 p.m. EDT about 250 miles above the south Pacific Ocean.

"Everything went very well," NASA mission commentator Kelly Humphries said during a televised broadcast of the docking.

Typically, the journey takes two days, but Russian engineers have developed new flight procedures that tweak the steering maneuvers and expedite the trip.

One other crew capsule and several cargo ships previously have taken the fast route to the station.

The express ride to the station began at 4:31 p.m. EDT when a Russian Soyuz rocket soared off its launch pad at the Baikonur Cosmodrome in Kazakhstan and deposited the crew's capsule into orbit. The spaceship circled around the planet less than four times before catching up to the station, a $100 billion project of 15 nations.

Overseeing operations from aboard the capsule was veteran cosmonaut Fyodor Yurchikhin, 54, who will be living aboard the station for the third time. The former commander also flew on NASA's now-retired space shuttle.

He was joined on the Soyuz by first-time astronaut Luca Parmitano, 36, a major in the Italian Air Force. Parmitano, who initially studied political science and international law at the University of Naples, is the first Italian to be assigned to a long-duration mission aboard the station, which is a laboratory for biomedical, materials science and other research.

"This is very momentous," Parmitano said in a preflight NASA interview.

NASA gave the crew slot to the Italian Space Agency as part of a barter agreement for Italian-made cargo haulers used during the shuttle program.

Rounding out the crew is U.S. astronaut Karen Nyberg, a 43-year-old mechanical engineer who has one previous spaceflight on her resume, a two-week shuttle mission. Back on Earth, her astronaut husband, Doug Hurley, is looking after their three-year-old son, Jack.

Nyberg, an avid quilter, said she was bringing along sewing supplies, a sketch book and pencils.

"I'm really hoping to spend some of my free time drawing," Nyberg said in a preflight interview. "I used to mostly draw portraits, and gave them to friends, but I haven't done it in a long time."

Awaiting their arrival were Russian station commander Pavel Vinogradov and flight engineers Alexander Misurkin, also a Russian, and Chris Cassidy, an American. The men are two months into a planned six-month mission.

The combined crews will oversee more than 100 research experiments and technology tests under way aboard the station. They also plan to conduct five spacewalks over the next three months, most of which are needed to prepare the station for a new Russian laboratory module due to arrive in December.

New gene delivery method: Magnetic nanoparticles

May 30, 2013 ? Stent angioplasty saves lives, but there often are side effects and complications related to the procedure, such as arterial restenosis and thrombosis. In the June 2013 issue of The FASEB Journal, however, scientists report that they have discovered a new nanoparticle gene delivery method that may overcome current limitations of gene therapy vectors and prevent complications associated with the stenting procedure. Specifically, this strategy uses stents as a platform for magnetically targeted gene delivery, where genes are moved to cells at arterial injury locations without causing unwanted side effects to other organs.

Additionally, magnetic nanoparticles developed and characterized in the study also protect genes and help them reach their target in active form, which also is one of the key challenges in any gene therapy.

"This study can help address a number of barriers to translation of experimental gene therapeutic approaches to clinical practice," said Michael Chorny, Ph.D., a researcher involved in the work from the Division of Cardiology at the Abramson Pediatric Research Center at The Children's Hospital of Philadelphia in Pennsylvania. "Bringing gene therapy closer to clinical use is a step toward developing safer and more effective ways for treating cardiovascular disease."

To make this technique possible, Chorny and colleagues used in vitro vascular cells to demonstrate the ability to effectively deliver genes using biocompatible nanoparticles and magnetic force without causing adverse effects. Although effective gene transfer in these cells has been difficult to achieve historically, this study demonstrated that magnetically guided "gene-impregnated" nanoparticles delivered their cargo effectively, especially compared to conventional gene delivery vectors. Next, researchers explored magnetically targeted gene delivery by applying these nanoparticles to stented arteries in rats. The nanoparticle-mediated expression of stent-targeted genes was shown to be greatly enhanced in treated animals when compared to control groups treated with nanoparticles without using the magnetic conditions, or with an equivalent dose of a conventional gene delivery vector. Genes delivered using the magnetically targeted nanoparticles were also expressed at considerably higher levels in the stented arteries compared to other organs and tissues.

"This approach is novel and exciting, and goes to show that investments in basic science across disciplines pay off in time," said Gerald Weissmann, M.D., Editor-in-Chief of The FASEB Journal. "When the first nanoparticles were developed and when the first correctable human disease gene was identified, no one could have ever known that these two advances would come together in a way that might one-day save lives."

It uses the retailer's existing trusted sign-in security and has already been tested on both Zappos and Woot, with both trials apparently noting "significant" pickup from customers. South Barrington homes are a magnet for luxury home buyers. The location is good, close to I-90 and Route 59. There are more luxury homes here and the typical cost of properties is higher than any other Barrington area. However I see a disconnect and I am not sure what can be done about it. We are talking about what a buyer wants and what?s available in terms of age of house.

New construction in South Barrington is very limited and before long likely will not exist at all. There is not a lot of land left that can be developed, and for now builders are not rushing to build spec homes for fear of not being able to sell them. Not many buyers are willing to buy a home from a rendition of what it might look like, they just don?t have the imagination.

The price ranch of $1 Million to $1.5 million will buy a home of about 5,000 square feet and this seems to be a target price and size for many of the buyers. It?s a fair statement that a buyer who wants to spend that amount of money and have a new home, would not get the size they want. There are limited ?homes in this price range in South Barrington but due to the age the buyers are having a hard time committing to buy them.

How can this disconnect be resolved?

  • South Barrington home buyers can wait the market for as long as they want until a house that is younger in age hits the market but who knows how long they?ll have to wait. They should also know they have competition. To think you are the only person looking for a younger house in South Barrington and make an offer based on that thought, will likely mean your offer will be trumped by someone else.
  • Dare I say look elsewhere? Likely not because South Barrington is known to be the best in 60010 for the luxury properties and location here is king!
  • Low ball offers until you score? Again this rarely works. Listing agents don?t just pluck numbers from the sky when they advise sellers what homes should sell for. We look at comparable sales. So sellers are not going to give in and sell because you try to get a cheap house. Knowing the market is more than knowing a price, it?s also about knowing market action, average market times, your competition.
  • Watch the mortgage interest rates. Absolutely. There has never been a time to get jumbo mortgages at such a low rate. Even this week we have seen the rates rise and this might affect your monthly outgoings more than you think. Lower payments could provide you the money you need to upgrade or change a house to be what you want it to be.
  • South Barrington home sellers cannot change the age of their homes but it might be worth updating the property to make it appeal to those buyers. Kitchens and bathrooms sell houses but windows, roofs and driveways are also big ticket items that buyers look at.

Either way there?s no way of making South Barrington?s housing stock any younger, buyers cannot tag older kitchens or windows as worthless, ?and something will have to give. With mortgage rates on the rise buyers may have to grasp hold of the fact that they cannot get all that they desire and take the plunge before prices start to rise. These limited choices are fully functioning and quite frankly beautiful homes,so why not update them over time. Sometimes you have to give to get, and an older kitchen in a superb waterfront location is an example.

Neuroscientists discover new phase of synaptic development

May 29, 2013 ? Students preparing for final exams might want to wait before pulling an all-night cram session -- at least as far as their neurons are concerned. Carnegie Mellon University neuroscientists have discovered a new intermediate phase in neuronal development during which repeated exposure to a stimulus shrinks synapses. The findings are published in the May 8 issue of the Journal of Neuroscience.

It's well known that synapses in the brain, the connections between neurons and other cells that allow for the transmission of information, grow when they're exposed to a stimulus. New research from the lab of Carnegie Mellon Associate Professor of Biological Sciences Alison L. Barth has shown that in the short term, synapses get even stronger than previously thought, but then quickly go through a transitional phase where they weaken.

"When you think of learning, you think that it's cumulative. We thought that synapses started small and then got bigger and bigger. This isn't the case," said Barth, who also is a member of the joint Carnegie Mellon/University of Pittsburgh Center for the Neural Basis of Cognition. "Based on our data, it seems like synapses that have recently been strengthened are peculiarly vulnerable -- more stimulation can actually wipe out the effects of learning.

"Psychologists know that for long-lasting memory, spaced training -- like studying for your classes after very lecture, all semester long -- is superior to cramming all night before the exam," Barth said. "This study shows why. Right after plasticity, synapses are almost fragile -- more training during this labile phases is actually counterproductive."

Previous research from Barth's lab established the biochemical mechanisms responsible for the strengthening of synapses in the neocortex, the part of the brain responsible for thought and language, but only measured the synapses after 24 hours. In the current study, post-doctoral student Jing A. Wen investigated how the synapses developed throughout the first 24 hours of exposure to a stimulus using a specialized transgenic mouse model created by Barth. The model senses its surroundings using only one whisker, which alters its ability to sense its environment and creates a sensory imbalance that increases plasticity in the brain. Since each whisker is linked to a specific area of the cortex, researchers can easily track neuronal changes.

Wen found that during this first day of learning, synapses go through three distinct phases. In the initiation phase, synaptic plasticity is spurred on by NMDA receptors. Over the next 12 hours or so, the synapses get stronger and stronger. As the stimulus is repeated, the NDMA receptors change their function and start to weaken the synapses in what the researchers have called the labile phase. After a few hours of weakening, another receptor, mGluR5, initiates a stabilization phase during which the synapses maintain their residual strength.

Furthermore, the researchers found that they could maintain the super-activated state found at the beginning of the labile phase by stopping the stimulus altogether or by injecting a glutamate receptor antagonist drug at an optimal time point. The findings are analogous to those seen in many psychological studies that use space training to improve memory.

"While synaptic changes can be long lasting, we've found that in this initial period there are a number of different things we could play with," Barth said. "The discovery of this labile phase suggests there are ways to control learning through the manipulation of the biochemical pathways that maintain memory."

Walking or bicycling to work influenced by others

May 29, 2013 ? People who walk or bike to work are likely to influence their co-workers and partners to do the same, according to health researchers.

"Social influences are important, specifically interpersonal influences, such as spouses and co-workers," said Melissa Bopp, assistant professor of kinesiology, Penn State. She emphasized that community and employers also significantly influence whether people choose to actively commute.

More than 80 percent of American adults do not meet the guidelines for aerobic and muscle-strengthening activities, according to Healthy People 2020, a federal initiative that sets national objectives and monitors progress concerning the health of the population. Regardless of a chronic disease or disability, any regular physical activity can improve health and quality of life. The U.S. Department of Health and Human Services recommends at least two and a half hours of moderate-intensity aerobic activity a week or one hour and 15 minutes of vigorous-intensity activity a week for adults.

Active commuting (AC) -- physical exercise, such as bicycling or walking, as a way to travel to and from work -- is one way to help adults integrate the recommended activity into their daily routine.

Bopp and colleagues report in the online issue of the American Journal of Health Behavior that married people were more likely to participate in AC than non-married people, men actively commuted more often than women and mothers were even less likely to actively commute.

Four of the variables studied probed the connection between interpersonal relationships and AC. Having a spouse who actively commutes or co-workers who actively commute had a positive influence on the decision to do the same. The perception that a spouse would approve of AC or that co-workers would approve of AC also had a positive influence, but with slightly less impact.

However, at an individual level variables that were negatively related to active commuting included age, BMI, number of children, number of chronic diseases and number of cars in the household.

Bopp noted she was surprised to discover how many variables were significantly related to active commuting. People who were comfortable with their bicycling skills were more likely to actively commute, as were those who believed they had a shorter biking or walking time to work. Believing that an employer supports active commuting and working for an employer who supports AC, living in a community that supports AC and believing that the community is supportive of pedestrians and bicyclists were all positively significantly related to active commuting.

On the opposite end of the spectrum, the researchers found that lack of on-street bike lanes, off-street bike and walking paths, and sidewalks all negatively influenced active commuting. Difficult terrain, bad weather and the speed and volume of traffic along the commuting route were also significantly related to people deciding to not actively commute.

The researchers distributed surveys to 9,766 people across the mid-Atlantic States and received 1,234 viable completed surveys. The respondents were between the ages of 18 and 75, employed full- or part-time and physically able to walk or bike to work. The participants responded to questions and statements including how they traveled to work, whether or not their spouse and coworkers influenced their choice on how they traveled to/from work, if their employer supported actively commuting, how confident they were with their cycling skills and how bicycle-friendly their community was.

The researchers did not include any questions about leisure-time activity or any other forms of physical activity participation in this study.

Moving forward, Bopp and colleagues believe that the findings of this study provide a foundation for large-scale strategies to target population-level AC patterns.

"We have to look at the complete picture and look at individual thoughts and beliefs (about AC)," said Bopp. "This is a complex problem that we need to think about at multiple levels to address influences on behavior."

Also working on this research were Andrew T. Kaczynski, assistant professor of health promotion, education and behavior, University of South Carolina, and Matthew E. Campbell, research assistant, Department of Kinesiology, Penn State.

A?fire?has been reported aboard a Royal Caribbean?cruise?ship?that was on its way to the Bahamas.

According to?cruise?line's statement on its website, the?fire?occurred on the mooring area of deck 3 about 2:15 a.m. Monday aboard the?ship?Grandeur of the Seas and was quickly extinguished.

Miami-based Royal Caribbean says the 2,224 passengers and 796 crew members had to report to evacuation stations but that all are safe and have been accounted for. The?cruise?line also says the?ship?still has full power. No major injuries were reported.

The?ship?left Baltimore on Friday for a 7-night?cruise. It was on its way to CocoCay in the Bahamas, but instead went to Freeport, Bahamas, for further evaluation.

It's a good thing there's plenty in theaters for people to see right now, because there isn't much coming out on home video this week. The only big release is a supernatural thriller from one of the people who brought you Paranormal Activity and Insidious. Then there are a handful of smaller releases that are, in fact, so small that you may not have heard of them. See below for this week's ultra-short list.

One of our favorite features of the Samsung Galaxy S4 is that it's essentially the same size as its predecessor, and so a number of accessories released for the Galaxy S3 work with the newer version as well. 

That brings us to what you see here -- the Samsung Multimedia Desktop Dock. It's a pretty basic deal -- plop your Galaxy S4 into it and it'll automatically switch to Desktop mode. Plug in with a microUSB cable and you'll charge the phone at the same time. And use the optional 3.5mm line out to play music while the phone's docked. 

This thing's pretty portable as well, with the dock section flipping down into itself for easy travel. And perhaps most important, you can get it fairly cheap since technically it's "last year's" dock.
